An HPLC-MS/MS method for the determination of trans-ferulic acid in human plasma

  • 摘要: 建立灵敏、快速的反式阿魏酸(trans-FA)血浆浓度的高效液相色谱-串联质谱(HPLC-MS/MS)分析方法,研究反式阿魏酸在健康中国受试者体内的药动学特征。血浆样品经乙酸乙酯液-液萃取,采用HederaODS-2色谱柱分离,流动相为甲醇-5 mmol·L-1乙酸铵水溶液(含0.05%乙酸) (34:66),流速为0.4 mL·min-1;质谱采用Turbo-Ionspray电喷雾离子源和负离子多反应监测方式。反式阿魏酸在0.1~5 ng·mL-1内线性关系良好(r≥0.9992);方法的日间和日内精密度均小于9.2%,准确度为95.4%~111.4%;无基质效应,无残留效应;稳定性良好。所建立的HPLC-MS/MS分析方法快速、灵敏、准确、专属性强、重现性高,适用于健康中国受试者体内反式阿魏酸的药代动力学研究。

     

    Abstract: In this study, we developed a sensitive and rapid HPLC-MS/MS method for the determination of trans-ferulic acid (trans-FA) in plasma samples, and investigated the pharmacokinetics characteristics in healthy volunteers. The plasma samples were extracted with acetic ether, and then separated on a Hedera ODS-2 column with a mobile phase of methanol and 5 mmol·L-1 ammonium acetate buffer solution containing 0.05% acetic acid (34:66) at a flow rate of 0.4 mL·min-1. Electrospray ionization source was applied and operated in the positive ion mode using MRM. The method exhibited a good linearity over the concentration range of 0.1-5 ng·mL-1 (r ≥ 0.9992). The values on both the occasions (intra-and inter-day) were all within 9.2%, and the accuracy was 95.4%-111.4%. No matrix effect and carry-over effect were observed. Trans-FA was stable in human plasma under different storage conditions. The developed HPLC-MS/MS method is rapid, sensitive, accurate, and reproducible, and suitable for the pharmacokinetic study of trans-FA in healthy Chinese volunteers.
