基于影像的脑胶质瘤原位移植瘤模型的建立

Establishment of a glioma orthotopic xenograft model based on imaging technology

  • 摘要: 脑肿瘤原位移植瘤模型是临床前评价抗脑瘤药物效果的有效手段。本研究利用荧光素酶和红色荧光蛋白双标记的人脑胶质瘤细胞U87MG-mCherry-luc,构建了基于影像的裸鼠脑原位移植瘤模型,利用已知的抗肿瘤药物替莫唑胺,结合小动物核磁共振成像系统(MRI)、活体成像系统(IVI)等影像技术验证了该模型的可行性。结果表明,该模型可有效地评价药物抗脑胶质瘤的效果。

     

    Abstract: Orthotopic xenograft model of human brain cancer cells is a good preclinical model for evaluation of antitumor compounds. In the present study, an orthotopic xenograft model of U87MG-mCherry-luc was established in Balb/c nude mice and the tumor growth was monitored via imaging technology including magnetic resonance imaging (MRI), in vivo imaging (IVI) and micro-CT. Furthermore, the model was evaluated with a positive drug temozolomide. Our data suggest that integrated imaging technology including MRI, IVI and micro-CT in orthotopic xenograft model can be used to facilitate monitoring of cancer progression and evaluate antitumor activity of drugs against glioma.
