肝脏和肠道酯酶在药物代谢及新药研发中的作用

Role of human liver and intestine esterases in drug design and development

  • 摘要: 人羧酸酯酶(human carboxylesterase,CES)和芳基乙酰胺脱乙酰酶(arylacetamide deacetylase,AADAC)是丝氨酸水解酶多基因家族的重要成员,不仅参与人体内源性胆固醇酯等的水解过程,还广泛参与药物在体内的代谢、活化和解毒过程,与酯类药物的个性化安全用药息息相关。本综述围绕人体内与药物代谢关系密切的酯酶,总结其结构及分布、代谢特点及近年来的研究进展,为新药研发和合理药物设计提供参考。

     

    Abstract: Human carboxylesterase (CES) and arylacetamide deacetylase (AADAC) are important numbers of the serine esterase superfamily. They are involved in hydrolytic procedure of human endogenous cholesteryl esters, as well as drug metabolism, activation and detoxication. They are closely related to the personalized medication of drugs, especially for prodrugs. This review summarizes their structure and distribution, metabolic characteristics and research progress in recent years, which will provide a reference for new drug development and rational drug design.
