波罗蜜根中的异戊烯基黄酮类成分

Prenylated flavonoids from the roots of Artocarpus heterophyllus

  • 摘要: 采用ODS、MCI、Sephadex LH-20、半制备HPLC等柱色谱技术对波罗蜜(桑科波罗蜜属)根的95%乙醇提取物进行了化学成分研究。从中分离得到了10个异戊烯基黄酮类成分,根据理化性质和波谱数据鉴定为artoheteroid E(1)、cycloheterophyllin(2)、artelastoxanthone(3)、artoindonesianin Q(4)、柘树黄酮C(5)、8-(γ,γ-dimethylallyl)-5,2',4'-trihydroxy-7-methoxyflavone(6)、桑皮酮T(7)、6-(3-methylbut-2-enyl)apigenin(8)、5,7,2',4'-四羟基-6-异戊烯基黄酮(9)和albanin A(10)。其中化合物1为新化合物,2~4为首次从该植物中分离得到。所有的化合物进行了抑制组织蛋白酶K(cathepsin K,CatK)活性筛选,其中化合物3~5、710对CatK具有不同程度的抑制作用,其IC50值分别为0.9、1.6、4.5、24.5、63.5μmol·L-1

     

    Abstract: Ten isoprenylated flavonoids were isolated from 95% ethanol extraction of Artocarpus heterophyllus, with a combination of various chromatographic approaches, including ODS, MCI, CHP-20 P, Sephadex LH-20 and high performance liquid chromatography. On the basis of physic-chemical characters and spectroscopic data analysis, these compounds were identified as artoheteroid E (1), cycloheterophyllin (2), artelastoxanthone (3), artoindonesianin Q (4), cudraflavone C (5), 8-(γ,γ-dimethylallyl)-5,2',4'-trihydroxy-7-methoxyflavone (6), kuwanon T (7), 6-(3-methylbut-2-enyl) apigenin (8), 5,7,2',4'-tetrahydroxy-6-(3-methylbut-3-enyl) flavone (9), albanin A (10). Among them, compound 1 is a new one, while compounds 2-4 were isolated for the first time from the plant of Artocarpus heterophyllus. All isolated compounds were screened for their inhibitory abilities against cathepsin K. Of them, compounds 3-5, 7 and 10 showed inhibitory effects with the IC50 values of 0.9, 1.6, 4.5, 24.5 and 63.5 μmol·L-1, respectively.
