蔓菁中两个新的三萜化合物及其抗癌活性研究

Anti-cancer activity of two new triterpenes from Brassica rapa

  • 摘要: 综合运用大孔吸附树脂、硅胶柱色谱、半制备型高效液相及Sephadex LH-20凝胶柱色谱等各种色谱技术,对蔓菁Brassicarapa L.乙醇提取物的化学成分进行系统研究,共分离得到4个三萜、3个黄酮和1个甾体类化合物。根据其理化性质和高分辨及二维核磁等波谱数据鉴定化合物的结构,其中12为新的三萜类化合物,37首次从该植物中分离得到。细胞毒活性显示12对5种常见的癌细胞均显示一定的抑制作用,尤其是对HL-60具有显著的细胞毒活性,其IC50分别为5.87和10.32 μmol·L-1

     

    Abstract: The chemical constituents from Brassica rapa were identified by various chromatographic techniques including silica gel, reversed-phase silica gel, macroporous resin and Sephadex LH-20 column chromatography. Eight compounds were isolated from this plant. The isolated compounds were elucidated by physicochemical properties and spectroscopic methods, including extensive 1D, 2D-NMR, and HR-ESI-MS techniques. Compounds 1, 2 are new triterpenoids and 3-7 were isolated for the first time from Brassica rapa. The cytotoxic effect of compounds 1 and 2 were tested by MTT assay against five cancer cell lines. The result showed that all compounds exhibit growth inhibition for the cancer lines. Compound 1 has an IC50 value of 5.87 μmol·L-1 for growth inhibition of leukemia cell line HL-60, and IC50 value for compound 2 was 10.32 μmol·L-1.
