UHPLC法测定经典名方清金化痰汤物质基准中多指标成分的含量

Simultaneous determination of multiple constituents of QingJinHuaTan Decoction by ultra high performance liquid chromatography

  • 摘要: 建立超高效液相色谱测定经典名方清金化痰汤物质基准中多指标成分含量的方法。采用Waters cortecs T3色谱柱(150 mm×2.1 mm,1.6 μm),以0.04%磷酸水(A)-乙腈(B)为流动相,进行梯度洗脱,流速为0.3 mL·min-1,柱温为25℃,DAD检测波长为238、280 nm。该色谱条件下,各成分色谱峰完全分离且峰形良好,各待测成分的线性关系良好(r>0.999),重复性、稳定性良好,回收率在92.5%~104.7%之间。本实验所建立的方法简便、准确、可靠,为清金化痰汤物质基准的质量评价提供了科学依据,为其开发和标准制定提供了参考。

     

    Abstract: A UHPLC method for the simultaneous determination of multiple constituents in QingJinHuaTan Decoction was established. The separation was performed on a Waters cortecs T3 column (150 mm×2.1 mm, 1.6 μm); the mobile phase was acetonitrile-water (containing 0.04% phosphoric acid) with gradient elution at a flow rate of 0.30 mL·min-1, the column temperature at 25℃ and the wavelengths at 238 nm and 280 nm. The results showed that all peaks were well separated and all components had a good linear relationship in the investigative range, (r > 0.999). The repeatability and stability were good and the recovery was between 92.5%-104.7%. The method is simple, accurate and reliable and provides a basis for quality control of QingJinHuaTan Decoction and for further development of methods for its standardization.
