长梗绞股蓝中的四个新达玛烷型三萜皂苷

Four new dammarane-type triterpenoid saponins from Gynostemma longipes C.Y.Wu

  • 摘要: 长梗绞股蓝(Gynostemma longipes C.Y.Wu)以达玛烷型三萜皂苷为主要活性成分。本文综合使用MCI柱色谱、硅胶柱色谱、制备型高效液相色谱等分离技术,从长梗绞股蓝中的大极性部位分离得到4个三萜皂苷类单体化合物,并主要利用MS和NMR等波谱学技术鉴定它们的结构为:(20S)-3β,20,21-三羟基达玛-19-醛基-24-烯-3-O-α-L-吡喃鼠李糖基(1→2)-β-D-吡喃木糖基(1→3)-α-L-吡喃阿拉伯糖基-21-O-β-D-吡喃葡萄糖基(1→6)-β-D-吡喃葡萄糖苷(1)、(20S)-3β,20,21-三羟基达玛-19-醛基24-烯-3-O-α-L-吡喃鼠李糖基-(1→2)-β-D-吡喃木糖基-(1→3)-α-L-吡喃阿拉伯糖基-21-O-α-L-吡喃鼠李糖基(1→6)-β-D-吡喃葡萄糖苷(2)、(20S)-3β,19,20,21-四羟基达玛-24-烯-3-O-α-L-吡喃鼠李糖基(1→2)-β-D-吡喃木糖基(1→3)-α-L-吡喃阿拉伯糖基-21-O-β-D-吡喃葡萄糖基(1→6)-β-D-吡喃葡萄糖苷(3)、(20S)-3β,20,21-三羟基达玛-24-烯-3-O-α-L-吡喃鼠李糖基(1→2)β-D-吡喃葡萄糖基(1→3)-β-D-吡喃葡萄糖基-21-O-β-D-吡喃葡萄糖基(1→6)-β-D-吡喃葡萄糖苷(4)。化合物14是四个新达玛烷型三萜皂苷,除了C-3位含有3个糖基外,还在C-21位均含有2个糖基。

     

    Abstract: Dammarane-type triterpenoid saponins are regarded as the main active constituents of Gynostemma longipes C.Y.Wu. By using MCI and silica gel column chromatography, as well as preparative HPLC, we isolated four new dammarane-type triterpenoid saponins from the polar saponin fraction of G. longipes C.Y.Wu. Their structures were determined by comprehensive analyses of NMR and MS data and identified as (20S)-3β,20,21-trihydroxydammar-19-oxo-24-ene-3-O-α-L-rhamnopyranosyl(1→2)-β-D-xylopyranosyl(1→3)-α-L-arabinopyranosyl-21-O-β-D-glucopyranosyl(1→6)-β-D-glucopyranoside (1), (20S)-3β,20,21-trihydroxydammar-24-ene-19-oxo-3-O-α-L-rhamnopyranosyl(1→2)-β-D-xylopyranosyl(1→3)-α-L-arabinopyranosyl-21-O-α-L-rhamnopyranosyl(1→6)-β-D-glucopyranoside (2), (20S)-3β,19,20,21-terahydroxydammar-24-ene-3-O-α-L-rhamnopyranosyl(1→2)-β-D-xylopyranosyl(1→3)-α-L-arabinopyranosyl-21-O-β-D-glucopyranosyl(1→6)-β-D-glucopyranoside (3), (20S)-3β,20,21-trihydroxydammar-24-ene-3-O-α-L-rhamnopyranosyl(1→2)-β-D-glucopyranosyl(1→3)-β-D-glucopyranosyl-21-O-β-D-glucopyranosyl(1→6)-β-D-glucopyranoside (4). Compounds 1-4 are new dammarane-type triterpenoid saponins which contain five glycosyl residues.
