定量系统药理学在抗肿瘤药物研究中的进展

Advances of quantitative systems pharmacology in antitumor drug research

  • 摘要: 肿瘤是全球死亡率最高的疾病之一。现有临床研究缺乏针对抗肿瘤药物疗效动态变化的定量描述,为给药方案的制订带来了困难。基于定量系统药理学(quantitative systems pharmacology,QSP)的研究,可通过整合肿瘤生长与体内分子、细胞之间的相互作用,定量考察药物与疾病之间的网络关系,预测抗肿瘤药物的疗效、毒性,探索效应机制及筛选生物标志物,为肿瘤患者的精准治疗提供新方法。本文介绍了QSP的定义、研究方法及在抗肿瘤药物中的典型案例,为QSP在抗肿瘤药物中的研发和应用提供参考。

     

    Abstract: The high mortality of tumor is one of the most urgent problems to be solved. However, the current clinical trials provide limited quantitative descriptions on dynamic changes of drug efficacy, which restrict the selection of dosing regimens. Quantitative systems pharmacology (QSP) is a new approach for precise treatment of tumors. It quantifies the network relationship between drug action and diseases by integrating the tumor growth and molecules, cells in vivo, thereby predicting the efficacy, toxicity, and mechanism of antitumor drugs as well as identifying predictive biomarkers. In this review, we provide an overview of definition of QSP, current approaches and typical applications in research of antitumor drugs to enhance our understanding of QSP.
