靶向流感病毒表面糖蛋白的小分子抑制剂研究进展

Recent advances in small-molecule inhibitors targeting influenza virus glycoproteins

  • 摘要: 血凝素和神经氨酸酶是流感病毒表面两种重要的糖蛋白, 分别在流感病毒生命周期的侵入和释放阶段发挥关键作用, 是药物设计的理想靶标。随着对流感病毒神经氨酸酶和血凝素的结构及功能的深入研究以及药物发现技术的不断发展, 针对这两个靶标的新型抑制剂被陆续发现。本综述精选近几年的代表性案例, 从药物化学角度总结了抗流感病毒表面糖蛋白小分子抑制剂的研究进展, 期望为新型高效抗流感病毒药物的研发提供参考。

     

    Abstract: Hemagglutinin and neuraminidase, two important glycoproteins on the surface of influenza virus, play a considerable role in the entry and release stage of the viral life cycle, respectively. With in-depth investigation of influenza virus glycoproteins and the continuous innovation of drug discovery strategies, a new generation of glycoproteins inhibitors have been continuously discovered. From the point of view of medicinal chemistry, this review summarizes the current advances in seeking small-molecule inhibitors targeting influenza virus glycoproteins, hoping to provide valuable guidance for future development of novel antiviral drugs.
