覃怀1号鲜地黄化学成分及其降糖活性研究

Chemical consitituents and hypoglycemic activity of Qinhuai No. 1 Rehmannia glutinosa

  • 摘要: 采用硅胶、MCI gel CHP-20、ODS、Toyopearl HW-40C等柱色谱技术, 并结合TLC和半制备液相等手段对覃怀1号鲜地黄70%丙酮提取物的乙酸乙酯部位进行分离纯化, 得到8个单体化合物。运用现代波谱学技术 (NMR、MS、UV、IR) 鉴定化合物的结构, 分别鉴定为地黄新苷A (1)、osmanthuside B6 (2)、地黄苷 (3)、异马蒂罗苷 (4)、反式对羟基肉桂酸 (5)、咖啡酸 (6)、阿魏酸 (7)、咖啡酸甲酯 (8)。其中, 化合物1为新苯乙醇苷, 鉴定为地黄新苷A, 化合物2为首次从地黄中分离得到。药理活性研究表明, 化合物267可以显著增加相对葡萄糖消耗量, 表现出潜在的降糖活性。

     

    Abstract: Eight compounds were isolated and purified from the ethyl acetate part of 70% acetone extract of Rehmannia glutinosa by various chromatographic techniques such as silica gel, MCI gel CHP-20, ODS, Toyopearl HW-40C, combined with TLC and semi-preparative HPLC. Their structures were elucidated by modern spectroscopy techniques (NMR, MS, UV, IR), and identified as neomartynoside A (1), osmanthuside B6 (2), martynoside (3), isomartynoside (4), (E)-p-hydroxycinnamic acid (5), caffeic acid (6), ferulic acid (7), and methyl caffeate (8). Compound 1 is a new phenylethanol glycoside, which was identified as neomartynoside A. Compound 2 was isolated from Rehmannia glutinosa for the first time. In addition, compounds 2, 6 and 7 significantly increased relative glucose consumption, showing potential hypoglycemic activity.
