基于中药超分子探究不同混合顺序对黄连解毒汤的物质形态及抗菌活性的影响

Effects of different mixing order on substance form and antibacterial activity of Huanglian Jiedu Decoction based on supramolecular Chinese medicine

  • 摘要: 中药超分子是中药复方的重要物质形态和显效形式, 超分子的形态与活性受多种因素影响, 混合顺序是其中关键要素之一。本研究采用动态光散射法、扫描电子显微镜、浊度仪及丁达尔效应等方法观察三组不同的混合顺序: 黄连组+栀子+黄芩(CGS)、黄连组+黄芩+栀子(CSG) 以及黄芩+栀子+黄连组(SGC) 汤剂之间的微观形态和物理性质差异, 利用高效液相色谱法分析其主要成分组成, 通过体外金黄色葡萄球菌(S. aureus) 模型对三组汤剂的抗菌活性进行初步对比, 并通过分子动力学模拟技术初步探讨三组汤剂中超分子的动态形成过程。研究表明, 不同混合顺序导致黄连解毒汤超分子部位形貌特征发生变化, 其宏观现象、电镜、粒径、电位、浊度等都伴随出现明显差异, 并显著影响其原有抗菌活性。此外, 分子动力学模拟结果也表明黄连解毒汤主要药效成分的不同混合顺序明显影响其自组装行为。本研究是对黄连解毒汤超分子物质基础形成的影响因素的进一步探索, 也为黄连解毒汤配方颗粒的临床合理服用提供了一定参考。

     

    Abstract: Chinese medicine supramolecular is an important material form and effective form of Chinese medicine compound. The form and activity of supramolecular are affected by many factors, and the mixing order is one of the key factors. In this study, dynamic light scattering (DLS), scanning electron microscopy (SEM), turbidimeter and Tyndall effect were used to observe the differences of micromorphology and physical properties of three different groups of Coptidis Rhizoma group + Gardeniae Fructus + Scutellariae Radix (CGS), Coptidis Rhizoma group + Scutellariae Radix + Gardeniae Fructus (CSG) and Scutellariae Radix + Gardeniae Fructus + Coptidis Rhizoma group (SGC). The main components of the three groups of decoction were analyzed by high performance liquid chromatography (HPLC). The antibacterial activity of the three groups of decoction was preliminarily compared by in vitro Staphylococcus model, and the dynamic formation process of supramolecules in the three groups of decoction was preliminarily discussed by molecular dynamics simulation (MDS) technology. The results showed that different mixing sequences led to changes in the morphology of supramolecular parts of Huanglian Jiedu Decoction, with significant differences in macroscopic phenomena, electron microscopy, particle size, potential and turbidity, and significantly affected the original antibacterial activity. In addition, the results of molecular dynamics simulation also show that the different mixing order of the main components of Huanglian Jiedu Decoction will significantly affect its self-assembly behavior. This study is to further explore the factors affecting the formation of supramolecular substance basis of Huanglian Jiedu Decoction, also provides a reference for the clinical rational administration of Huanglian Jiedu Decoction formula granules.
