Advances in chemically synthesized glycopeptide-based antitumor vaccines

  • 摘要: 在肿瘤发生过程中, 糖基转移酶的表达会发生显著变化, 这导致与健康细胞相比, 肿瘤细胞表面的糖蛋白呈现出独特的糖链结构特征, 即高度截短的糖链。此类异常糖基化的产物被称为肿瘤相关糖抗原(tumor-associated carbone antigens, TACAs)。作为恶性肿瘤的重要分子标志物, TACAs不仅参与调控肿瘤的发生、发展、侵袭和转移等多个关键环节, 同时在肿瘤免疫逃逸中发挥重要作用。基于此, 靶向肿瘤的异常糖基化蛋白质或肽段已经成为目前抗肿瘤药物研发的重要策略之一。然而, 从天然来源中分离糖蛋白较为困难, 且使用化学方法合成具有复杂结构的糖蛋白或者糖肽仍具有一定的挑战性。本文系统综述了近年来利用化学方法合成糖肽抗肿瘤疫苗的研究进展, 以期为后续糖肽抗肿瘤疫苗的研发提供理论依据和技术参考。

     

    Abstract: There exist significant changes in glycosyltransferase expression during tumorigenesis, which leads to the unique sugar chain structural characteristics of glycoproteins on the surface of tumor cells compared to normal cells, namely highly truncated sugar chains. These aberrant glycosylated products are called tumor-associated carbohydrate antigens (tumor-associated carbone antigens, TACAs). As an important molecular marker of malignant tumors, TACAs not only participate in the regulation of tumor occurrence, development, invasion and metastasis of tumors, but also play a key role in tumor immune escape. Thus, targeting abnormally glycosylated proteins or peptides on tumors have become one of most important anti-tumor drug development strategies. However, it is difficult to isolate glycoproteins from natural sources, and it is still challenging to synthesize complex glycoproteins or glycopeptides by chemical methods. This review summarizes the progress of glycopeptide anti-tumor vaccine synthesized by chemical methods in recent years, in order to provide theoretical basis and technical reference for the research and development of glycopeptide anti-tumor vaccine in the future.
