基于电子舌技术的药物掩味效率多设备通用评价方法研究

Research on multi-equipment general evaluation method of drug taste-masking efficiency based on electronic tongue technology

  • 摘要: 药物不良味道掩蔽水平(掩味效率)直接影响药物适口性, 进而影响服药依从性, 但目前尚无有效、统一的评价方法。本文选取盐酸非索非那定干混悬剂、碳酸钙D3颗粒、复方聚乙二醇电解质散这3种不同剂型、不同溶解性的药物, 在TS-5000Z、Astree Ⅱ V5、iTongue20这3款电子舌上同步开展测试研究, 通过溶液合理制备、传感器有效清洗和样品响应可重复的判断准则的确定, 结合主成分分析、欧氏距离等高维数据相似度判别方法, 建立一种以参照溶液为基准、以样品溶液与参照溶液间滋味相似程度为标尺的药物掩味效率评价方法, 获得了结果可重现、仪器间可比较且与人尝结果一致的测试方法。上述方法是经标准化的且可多类型药物以及多原理设备通用的新方法, 为药物制剂间口味适宜程度的比较提供技术支撑。

     

    Abstract: The masking level of adverse taste of drugs, defined as taste-masking efficiency, directly affects the palatability of drugs, which in turn affects medication compliance. However, there is no effective and unified evaluation method. In this paper, three preparations with different dosage form and varied solubility, fexofenadine hydrochloride dry suspension, calcium carbonate D3 particles and compound polyethylene glycol electrolyte powder, were selected to carry out simultaneous research on TS-5000Z, Astree Ⅱ V5 and iTongue20 electronic tongues. Through the determination of judgment criteria for reasonable solution preparation, effective sensor cleaning and repeatable sample testing, combined with high-dimensional data similarity discrimination methods such as principal component analysis and Euclidean distance, an evaluation method of taste-masking efficiency for drugs was established based on the design of reference solutions and determination of taste similarity between sample solution and reference solution. The above method was reproducible with comparable results among instruments, and with consistent results with human sensory evaluation. The above method is a standardized new method that can be used for multi-type drugs and multi-principle equipment, which provides technical support for the comparison of taste palatability between pharmaceutical preparations.
