灯盏花素在家犬体内的药代动力学

Study on the pharmacokinetics of scutellarin in dogs

  • 摘要: 目的建立高效液相色谱法测定家犬血浆中灯盏花素主要有效成分灯盏乙素的浓度,研究灯盏花素在家犬体内的药代动力学。方法用高效液相色谱法测定6只家犬iv灯盏花素(以灯盏乙素计为120 mg/只)后不同时间血浆中灯盏乙素的浓度,绘制药-时曲线,计算药代动力学参数。结果灯盏乙素的药-时曲线符合三室模型,其T1/2 γ,T1/2α和T1/2β分别为(1.1±0.8) min,(7.0±2.8) min和(52±29) min;Vc为(880±508) mL;CL为(190±54) mL·min-1;AUC0-90和AUC0-∞分别为(574±134) mg·min·L-1和(599±132) mg·min·L-1。结论灯盏花素家犬iv给药后,血浆中灯盏乙素浓度迅速下降,提示制剂开发的剂型选择、临床给药方法或给药间隔时间的确定都应该考虑其T1/2

     

    Abstract: AimTo determine scutellarin in dog plasma and study the pharmacokinetics of scutellarin in the dog. MethodsScutellarin in plasma of six dogs at different sampling time was determined after single dose of 120 mg iv by RP-HPLC. The mean plasma concentration-time curve was protracted and pharmacokinetic parameters were calculated. ResultsThe concentration-time curve of scutellarin can be fitted to a three-compartment model with the main pharmacokinetic parameters as follows: T1/2 γ, T1/2α and T1/2β were (1.1±0.8) min, (7.0±2.8) min and (52±29) min, respectively, Vc was (880±508) mL, CL was (190±54) mL·min-1, AUC0-90 and AUC0-∞ were (574±134) mg·min·L-1 and (599±132) mg·min·L-1 respectively. ConclusionThe concentration of scutellarin in plasma declined rapidly after single dose of 120 mg iv in dogs, and this suggested that the T1/2 of scutellarin should be taken into account in preparation exploitation and drug administration.
