南蛇藤素抑制豚鼠体外精子的受精能力

IN VITRO INHIBITION OF CELASTROL ON SPERMATOZOA FERTILIZATION ABILITY OF GUINEA PIG

  • 摘要: 南蛇藤素(Cel)对豚鼠精子前向运动(FM)、获能(Cap)、顶体反应(AR)和穿透去透明带仓鼠卵(SPA)均有明显的抑制作用,其作用强度随剂量而增加;对Cel的敏感性依次为精子Cap>FM>SPA>AR。Cel对豚鼠精子AR,FM和Cap的抑制作用比乙酸棉酚(GA)明显为强。

     

    Abstract: The effects of celastrol(Cel),isolated from Tripterygium wilfordii,on guinea pigsperm forward motility(FM),capacitation(Cap),the acrosome reaction(AR)and spermpenetration assay(SPA)were assessed in vitro.Cel(5 μg· ml-1)was found to inhibit thesespermatozoal functions,and the inhibitions were proportional to the concentrations of Cel used.Thepotency of inhibition of Cel on the fertilizing ability in guinea pig spermatozoa in vitro seems to followthe order :Cap>FM>SPA>AR.The inhibitory effect appeared to be reversible after washing awayCel if the duration of exposure of spermatozoa to Cel was shorter than 3 h. In a comparative study,the inhibitory effects of Cel on guinea pig sperm FM and AR were significantly stronger than those ofgossypol acetic acid.
