三尖杉酯碱类的高速液相色谱法分离与测定

HPLC SEPARATION AND DETERMINATION OF CEPHALOTAXUS ALKALOIDS

  • 摘要: 本文报道以Lichrosorb SI-60及Nucleosil C-18为固定相,用正相和反相高速液相色谱法分离三尖杉总生物碱、混合酯碱、及半合成三尖杉酯碱差向异构化合物,并用于混合酯碱、半合成三尖杉酯碱差向异构化合物的含量测定,比较了各种参数变化对色谱过程的影响,以拟定的方法对各种酯碱提取物、制剂及半合成产品作了定量分析。本法的灵敏度为1×10-6g,平均偏差1.9%。

     

    Abstract: Normal and reverse phase HPLC methods using Lichrosorb SI-60 and Nucleosil C-18 as the stationary phase in separation of total alkaloid of Cephalotaxus, mixed alkaloid esters and epimers of partially synthesized Cephalotaxus alkaloids are reported. The method proposed can also be used for the quantitative analysis of mixed alkaloid esters and epimers of partially synthesized Cephalotaxus alkaloids. The influence of analytical parameters upon the chromatographic behaviour has been discussed. The methods suggested were found to be suitable for the analysis of total alkaloid extract, preparation and semi-synthesized products. The sensitivity of the method is 1× 10-6 g with average deviation of 1.9%.
