川芎嗪在兔体内代谢产物的研究

STUDIES ON THE METABOLITES OF TETRAMETHYLPYRAZINE IN RABBITS

  • 摘要: 应用高效液相色谱法将川芎嗪在家兔血液中的代谢产物进行分离,对原药及两种代谢产物进行了紫外、红外、质谱、核磁共振等光谱分析,推断出川芎嗪代谢产物的结构,并用化学方法合成了代谢物对照品,对其结构进一步得到验证。

     

    Abstract: The metabolites of tetramethylpyrazine in rabbit have been separated by HPLC and solvent extraction. Their structures were identified by means of UV, IR, MS and NMR spectroscopy. The possible metabolic ways of TMPz are oxidization of one of the four methyl groups to hydroxymethyl and carboxyl group forming 2-hydroxymethyl-3,5,6-trimethylpyrazineand 3,5,6-trimethylpyrazine-2-carboxylic acid . These were further confirmed by the identity of HPLC data and all the above spectral parameters of the metabolites in vivo and the compounds objectively synthesized from TMPz.
