氯霉素含量测定法的研究 (一)比色分析法
METHODS FOR THE DETERMINATION OF CHLORAMPHENICOL(Ⅰ)COLORIMETRIC METHOD

摘要: 1.本法叙选了一种氯霉素的此色含量测定法,达方法是以Peppe 氏法为基础而加以改良,即将其还原剂氯化亚锡改为锌粉,使硝基还原为氨基,重氮化后与α-萘酚偶合,而省去了用乙醚抽提等手续。2.本法适用于氯霉素左旋体或混旋体及其在一般制剂中的含量测定。测定时氯每素的浓度以调整至0.13—0.3毫克/10毫升最为适宜。偶合时α-萘酚用量必须一致,在上述氯霉素浓度下,以加入1%的醇溶液0.1毫升为宜。显色所需时间为30分钟。锌盐在比色时无影响。Abstract: A colorimetric method for the determination of chloramphenicol is described. A sample containing 10 mg of chloramphenicol was reduced with 10 ml of 10% hydrochloric acid arid 50 to 100 mg of zinc dust,diluted with water to 500 ml, of which 10 ml was diazotized with 0.5 ml of 10% solution of sodium nitrite, then coupled with a mixture of 10 ml of 5% sodium hydroxide solution and 0.1 ml of a solution of 1% α-naphthol in alcohol for 30 minutes.It was then diluted to 100 ml.The color developed was found to be stable.The optical density was then taken from a B.Lunge's Model Ⅳ Universal Colorimeter using a BG-7 filter (480 mμ),and the result read off from a standard curve previously prepared from a purified sample of racemic chloramphenicol manufactured in our own works. Suitable concentration of chloramphenicol for comparison was found to be between 0.13 and 0.3 mg/10ml.The amount of α-naphthol used was found to influence the result.
