UPLC-MS/MS法测定注射自微乳大鼠体内熊果酸血药浓度及其药动学研究

Plasma concentration and pharmacokinetics of ursolic acid carried in self-microemulsifying drug delivery system in rats studied by UPLC-MS/MS

  • 摘要: 建立注射自微乳大鼠体内熊果酸血药浓度的超高效液相色谱-串联质谱测定方法,并研究其在大鼠 体内的药动学。血清中加入内标并经C18固相小柱萃取后进行测定分析。采用Acquity UPLC BEH C18色谱柱(100 mm × 2.1 mm,1.7 μm)分离;流动相: 乙腈(A)- 0.1% 氨水(B),梯度洗脱;流速: 0.2 mL·min-1,检测波长210 nm,柱温40 ℃;采用电喷雾离子源(ESI)以负离子方式检测;扫描方式为多反应离子监测方式(MRM)检测。熊果酸血药浓度在1.19~3 815.00 ng·mL-1内线性关系良好(r= 0.999 0);不同浓度的提取回收率在87.42%~89.95% 之间;日内、日间精密度均小于11%。建立的方法灵敏度高、简单快速,可用于大鼠血清中熊果酸的药动学研究。

     

    Abstract: This study is to report the establishment of an UPLC-MS/MS method for the determinationof plasma concentration of UA carried in self-microemulsifying drug delivery system (SMEDDS) and its pharmacokinetics in rats.It was used for determination and analysis when serum with internal standard was extracted from C18 solid-phase column.Acquity UPLC BEH C18 column (100 mm × 2.1 mm, 1.7 μm) was used for separation.The mobile phase was acetonitrile - 0.1% ammonia with gradient elution at the flow rate of0.2 mL·min-1.The column temperature was 40 ℃ and the detection wave length was 210 nm.It was detected by negative ion using electrospray ionization source (ESI) and scanned by multiple reaction ion monitoring (MRM) mode.The liner relationship of UA was very good in the range of 1.19 - 3 815.00 ng·mL-1 (r = 0.999 0).Recovery rate of different concentrations were 87.42% - 89.95%.The precision of inter-day and intra-day were less than 11%.The method developed in our study was proved to be sensitive, rapid and simple.It is suitable for the pharmacokinetic study of UA-SMEDDS in rats.
