斑蝥酸钠在小鼠体内抗癌作用的研究

STUDIES ON THE ANTINEOPLASTIC ACTION OF THE DISODIUM SALT CANTHARIDIC ACID IN THE MOUSE

  • 摘要: 斑螯Mylabris spp.体内抗癌有效成分为斑蝥素及斑蝥酸钠,已经北京及上海协作区五十多个医院临床试验肯定了效果。本文报告了3H-斑螯酸钠在艾氏腹水癌小鼠体内的吸收、分布和代谢。血液中的放射性代谢速率,以2小时最高,24小时在血中尚存留很少量,72小时测不到放射性。3H-斑蝥酸钠在组织中的代谢是4小时进入脑、胆、肾、肝四种组织的量最多,其他组织较少。3H-斑蝥酸钠用电镜放射自显影方法研究其对癌细胞的作用,曝光40天观察到3H-斑蝥酸钠进入细胞线粒体较多,由核孔进入细胞核及核仁也很多;对照组则没有。本实验进一步验证了临床的效果。

     

    Abstract: This paper deals with the distribution of the disodium salt of cantharidic acid in mice transplanted with Ehrlich carcinoma. Each mouse was given 0.133/μc/ml of 3H disodium salt of cantharidic acid intraperitoneally, and killed 1, 4, 24, and 48 hrs. after injection.The cancer cells were collected from the animal, fixed in 4% glutaraldehyde and osmium tetroxide, then kept in 1/15 M phosphate buffer at pH 7.2—7.4. After centrifugation, dehydration and embedding in Epon 812, samples were cut into ultrathin sections with a LKB ultratome, double stained with leadcitrate and uranyl acetate, and observed with a DX-2 electron microscope. Autoradiographic procedures were carried out with Ilford L4 single layer emulsion and exposed for 40—90 days at 4℃.The radioactivity of the tritium labeled compound in the blood and tissues were measured with a liquid scintillation counter. It was found that the radioactivity of the blood sample reached its maximum 2 hrs. after administration. According to the decrease of their radioactivity, the tissues were ,arranged in sequence as follows: brain, gall bladder, kidney, liver, thymus, spleen, lung, heart, stomach, mus.c.le, etc, with. brain being the highest and intestine the lowest.With electron microscopic autoradiography it was shown that the labeled compound located mainly in the nuclei and mitochondria of the cancer cells.
