二氢青蒿素的变旋作用与差向异构体转化研究

STUDIES ON MUTAROTATION AND ANOMERIC CONVERSION OF DIHYDROQINGHAOSU

  • 摘要: 二氢青蒿素在极性溶媒中,其旋光度随时间而变化,可达一恒定值。高效液相色谱测定表明,在溶液中,该物质有一自β差向异构体向α型转化的动力学过程,此过程与旋光度的变化相对应,其转化速率常数随溶剂、温度而异,酸具催化作用。这一现象的本质是半缩醛的变旋作用。

     

    Abstract: Polarimetry and HPLC were used to study the solution of dihydroqinghaosu in polar solvents such as methanol and ethanol. The mutarotation and conversion of anomers of dihydroqinghaosu were observed and a few kinetic parameters were measured. It was found that the equilibrium constant and the initial forward rate constant of anomeric conversion of dihydroqinghasu (βtoα) depended upon the solvents, temperature and other factors. Hydrochloric acid can be used as a eatalyst for the conversion reaction. A mechanism of the mutarotation and anomeric conversion of dihvdroqinghaosu a kind of hemiacetal was peoposed.
