HPLC SEPARATION AND DETERMINATION OF ACTIVE CONSTITUENTS IN CHIN PIE (FRAXINUS CHINENSIS)

  • 摘要: 本文报道用高效液相层析法分离和测定秦皮乙醇提取液中的有效成分。填充剂为十八烷基键合相,洗脱剂为甲醇—水—乙酸(31:69:0.4)。七叶亭、七叶灵、梣皮甙和丁香甙得到基线分离。用254nm检测器检测,测定结果用数据处理机处理和计算。方法简便、灵敏、结果准确(变异系数为2.4%),分析三种生药样品,得出了结果。

     

    Abstract: In this paper the chromatographic behaviors of esculin, syringin, fraxin and esculetin using reversed phase high performance liquid chromatography have been studied. These compounds in ethanolic extract of Chin Pie can be well separated on an ODS column (25cm×5mm) with methanol-water-acetic acid (31:69:0.4) as eluant, UV-254 nm detector was used. Linear working curves prepared from peak areas vs. weights of the standards were obtained for the four mentioned constituents.The proposed method is simple; rapid and more sensitive than the TLCdensitometric method. The coefficient of variation was 2.4%, The analytical results were in good agreement.
