简捷实用的半数致死量综合计算法

  • 摘要: 本文根据适当的实驗設計即:(1)各組动物数相同;(2)剂量成等比級数;(3)反应情况大致符合常态分布;(4)实驗例包括或接近0%及100%反应率,簡化并改进了Kaerber法,又結合了Bliss几率单位法的特点,提出一种甚为簡捷而又相当准确的半数致死量計算法。文中通过数理論証和实例校算証明了本法在簡捷度上优于Reed-Muench法,Kaerber原法及:Thompson法。在精确度上亦較Kaerbor原法有所进步。計算結果与Bliss正規法很为接近。主要公式为: logLD50=Xm-i(∑p-0.5) SE50=i×((∑p-∑+(p2))/(n-1))1/2 b=(Yh-Yt)/(i(N-H)) log LDK=log LD50-(5-YK)/b SEK=((SE50)2+2/(nH(N-H))((5-YK)/b2i)2)1/2 自1927年Trevan提出半数致死量(LD50)以来,因其較高的精确性和重复稳定性受到普遍重視。近年来,在药物篩选、毒性研究、安全系数及效价測定等方面,半数致死量LD50及其有关数据(LD5,LD10,…b,ED50)等应用日益广泛。通过Behtens、Kaerber、Bliss、Reed及Muench、van der Waerden、Litchfield及Wilcoxon,Thompson,顾汉頤等的研究,已出現不少計算方法。或以簡捷見长,或以精确取先,或在一定实驗設計下将精确但繁复的計算法进行簡化以求实用。本文在合理的实驗安排下,簡化和改进了Kaerber法,并綜合了Bliss法的特长,提出一种甚为簡捷而又相当精确的綜合計算法。在药理学、生物学、病毒学等方面具有一定的实用意义。

     

    Abstract: A simpler and reasonably accurate method for computing the median lethal dose (LD50 is described. The computing process is much simpler than Reed-Muench's method, and the accuracy attainable is precisely the same as that of Kaerber's method. By using this method, one can easily obtain the regression equation correlating the log-dose and the "probit" unit, and thereby compute the dosage for any "percentage kill", without the necessity of repeated correction. The method is also free from subjective influence.
