人参皂苷Rg1及其肠内菌代谢产物Rh1对小鼠免疫细胞功能的影响

EFFECTS OF GINSENOSIDE Rg1 AND ITS METABOLITE Rh1 ON THE FUNCTION OF IMMUNOCYTE IN MICE

  • 摘要: 目的初探人参皂苷Rg1及其肠内菌代谢产物Rh1对正常小鼠免疫功能的影响。方法用Rh1与Rg1分别处理脾T细胞,B细胞及腹腔巨噬细胞(Mφ);MTT比色法测T和B细胞增殖能力;中性红比色法测Mφ的吞噬功能;Griess法测Mφ释放NO的水平。结果Rh1能促进脾细胞增殖、下调Con A诱导的T细胞增殖;Rh1与Rg1对LPS诱导的B细胞增殖均无明显作用;Rg1和Rh1能提高Mφ的吞噬能力和促进NO的释放。结论Rg1及其代谢产物Rh1可共同作用于T细胞和Mφ而产生免疫调节作用。

     

    Abstract: AIMTo study the mechanism of action of ginsenoside Rg1 and its metabolite Rh1 on the function of immunocyte in normal mice. METHODSThe proliferation of splenocytes was determined by using MTT assay. Phagocytosis activity of peritoneal macrophages was determined by using neutral red phagocytosis assay. Griess agent was used to assay the NO-releasing level of peritoneal macrophages. RESULTSRh1 was shown to promote proliferation of splenocytes directly, down-regulate the Con A-induced proliferation of T-lymphocytes. Rg1 can not induce proliferation of splenocytes directly, while Rg1 100 μg·mL-1 can inhibit ConA-induced proliferation of T-lymphocytes obviously. Both Rh1 and Rg1 showed no effect on LPS-induced proliferation of B-lymphocytes. Rg1 and Rh1 were found to significantly increase the phagocytosis and NO-releasing level of peritoneal macrophages, while the effect of Rh1 is weaker than that of Rg1. CONCLUSIONThe immuno-regulation effect of Rg1 may be carried out by both Rg1 and its metabolite Rh1. Their functions on other immunocytes still need further study.
