A-环失碳19-去甲基双炔甾族化合物的合成

THE SYNTHESIS OF A-NOR-19-NOR-DIETHYNYL STEROIDS

  • 摘要: 本文报道利用消旋18-甲基炔诺酮的中间体17β-羟基19-去甲基13β-乙基-4-雄甾烯-3-酮(Ⅲ)和炔诺酮的中间体19-去甲基-4-雄甾烯-3,17-双酮(Ⅵ),经锂氨还原,氧化、环合、炔化四步反应合成A-环失碳19-去甲基2,17α-双乙炔基5α-雄甾烷2,17β-双丙酸酯(Ⅻb)及其18-甲基类似物(Ⅻa)。A-环失碳双炔甾族化合物-抗孕53号具有较弱的雌激素样活性,且在临床上有一定的抗生育效果,毒性较低。根据10-位和13-位取代基对甾体激素生物活性的影响,我们改变抗孕-53号分子中该两位的取代基。合成了目的物(Ⅻa)和(Ⅻb)。

     

    Abstract: The synthesis of a potential anti-implanting agents for female, A-nor-19-nor-2, 17 α-diethynyl-5 α-androstane-2,17 β-diol dipropionate and its 18-methyl analog featured with a shortened approach was reported. 19-nor-4-androsten-3,17-dione and 17 β-hydr-oxy-19-nor-13 β-ethyl-4-androsten-3-one were used as the starting materials, respectively and then followed successfully with lithium ammonia reduction, oxidation, cyclization. and final ethynylation to afford the desired compounds.
