STUDIES ON STABILIZATION OF DRUGS IN SOLUTION Ⅲ. A CHEMICAL KINETIC STUDY OF OXACILLIN IN AQUEOUS SOLUTION

  • 摘要: 苯甲异噁唑青霉素钠在水溶液中的降解为伪一级反应,受H+,OH-的催化,HPO4-的催化也相当剧烈.20℃时最稳定的pH值为6.54.在35℃,pH 1.5时,水溶液较青霉素G稳定9-10倍.37℃时在pH 6.5的一倍稀释的McIlvaine缓冲溶液中较青霉素G稳定1.5倍.0℃储藏十分稳定.文中给出了各种速度常数如KHA-,KOHA-,Ko,KHPO4-的Arrhenius公式和稳定性的预测数值.

     

    Abstract: A chemical kinetic study of the stability of oxacillin in aqueous solution at 3 dif- ferent temperatures (35℃, 40℃ and 50℃) and with a pH range from 1.44-10.93 was carried out. Concentrations were determind by the iodometric method, the latter being proved by the authors to be well in accord with the microbiological method. The degradation of oxacillin was proved to be a first-order reaction at any given pH. The apparent first-order constant K is KHA- H+ + KOHA-OH- + Ko + KHPO4=HPO4= . The activation energy Ea and frequency factors are listed in Table 3. The t1/2 and t9/10 (hr) of oxacillin from pH 4.5 to 8.0 at 0℃ and 20℃ are presented in Table 5, where column a is for the solutions without HPO4= ion catalysis and other general acid-base catalysis, and column b for those solutions made by 1/2 strength of the McIlvaine buffer system.
