金银花苯丙氨酸解氨酶(LJPAL1)抗体制备及酶联免疫吸附分析

Antibody preparation and enzyme linked immunosorbent assay of Lonicera japonica phenylalnine ammonia lyase

  • 摘要: 本文基于已有工作基础, 选择与金银花活性成分积累密切相关的苯丙氨酸解氨酶基因LJPAL1构建 原核表达载体, 并在大肠杆菌中进行诱导表达; 利用3个抗原位点分别合成多肽抗原, 并制备多克隆抗体Anti- LJT-1、Anti-LJT-2和Anti-LJT-3; 利用蛋白质印迹法筛选抗体Anti-LJT-2, 初步建立了间接法ELISA。本研究结果为进一步开展金银花化学质量快速评价试剂盒研究奠定基础。

     

    Abstract: The expression of phenylalnine ammonia lyase (LJPAL1) is closely related to the content of active compounds in Lonicera japonica. In this paper, a prokaryotic expression vector is constructed and LJPAL1 protein is expressed in E. coli. Three antigen sites were synthesized to peptide antigen and prepared polyclonal antibody of Anti-LJT-1, Anti-LJT-2 and Anti-LJT-3, separately. Antibody Anti-LJT-2 was screened using Western blotting. And indirect ELISA was built using Anti-LJT-2. The results of this study will be a base for honeysuckle chemical quality and evaluation kits.
