单纯疱疹病毒对常用抗病毒药物的耐药性

DEVELOPMENT OF RESISTANCE OF HERPES SIMPLES VIRUS TO ANTIVIRUS DRUGS IN VITRO

  • 摘要: 单纯疱疹病毒(HSV)与药物共同培养,传数代后,对单一抗病毒药物无环鸟苷(ACV)、环胞苷(CC)、丙氧鸟苷(DHPG)和膦羧基甲酸钠(PFA)均可产生耐药性。但耐药性产生的时间和程度各有不同。联合用药可延缓并减少病毒耐药株的发生,同时可降低用药浓度和剂量,从而减少毒性反应。不同耐药株的交叉药敏试验结果为临床合理用药提供了实验依据。

     

    Abstract: Resistant variants of HSV-1 wt stain to ACV, CC, DHPG and PFAcan be selected after several passages in drug-containing cultures. The degree of resistanceof different variants varied and emerged at different times. The combination of antiviralagents can delay and even prevent the emergance of drug-resistant variants. At the sametime, the concentration, dose and, therefore, toxicity of the antiviral agents can also be re-duced. So, it is a feasible method for treating resistant HSV infections. Cross-sensitivitytests showed that ACVr variants are resistant to ACV, DHPG and PFA, but sensitive toCC; PFA~r variants are sensitive to ACV, CC and DHPG; CCr variants are sensitive toACV and DHPG, but resistant to PFA; DHPGr variants are sensitive to ACV, CC andPFA, These results suggest that the study of cross-sensitivity of HSV strains in vitro pro- vide information which will aid the design of sultable therapy for drug-resistant HSV in-fections.
