Determination of chlorogenic acid and eriodictyol-7-O-β-D-glucuronide in Pyrrosia by RP-HPLC

  • 摘要: 目的 对中药石韦中的2种成分绿原酸和圣草酚7-O-β-D-吡喃葡糖醛酸苷的分析方法进行研究,并测定了不同种、不同产地石韦中2种成分的含量。方法 用反相C18柱,甲醇-水-磷酸(50∶200∶0.2)为流动相,284 nm作为检测波长。结果 绿原酸和圣草酚7-O-β-D-吡喃葡糖醛酸苷的含量分别在0.01~5 μg和0.004~5 μg有良好的线性关系(R=0.999 7),样品的回收率分别为97.9%(RSD=2.9%)和99.1%(RSD=2.4%)。结论 采用RP-HPLC法对21个不同种、不同产地石韦样品进行了测定,不同产地、不同种石韦中绿原酸及圣草酚7-O-β-D-吡喃葡糖醛酸苷的含量相差悬殊,本方法为控制中药石韦质量及合理采收提供了简便易行的方法。

     

    Abstract: Aim To develop a method for determination of chlorogenic acid and eriodictyol-7-O-β-D-glucuronide in Pyrrosia of different species and different places of origin by RP-HPLC. Methods Chromatography was performed using a C18 column with mobile phase of methanol-water-phosphoric acid (50∶200∶0.2). The monitoring wavelength was 284 nm. Results The linear ranges were 0.01-5.0 μg (R=0.999 7) and 0.004-5.0 μg (R=0.999 7), the recoveries were 97.1% (N=8, RSD=2.7%) and 98.8% (N=9, RSD=2.5%) for chlorogenic acid and eriodictyol-7-O-β-D-glucuronide, respectively. Conclusion The method was employed to the analysis of 21 samples of Pyrrosia. The contents of compounds vary greatly depending on the species used, place of collection and time of harvesting. The HPLC method is sensitive, rapid and can be used to control the quality of Pyrrosia and to guide reasonable season of harvesting.
