高效液相色谱法测定血浆中氟卡胺浓度及其在人体内的药代动力学
DETERMINATION OF FLECAINIDE CONCENTRATION IN BLOOD PLASMA BY HPLC AND PHARMACOKINETIC PARAMETERS IN VOLUNTEERS

摘要: 本文报告了氟卡胺血药浓度的高效液相色谱测定法。以氟基安定为内标,以含有离子对试剂的甲醇—水溶液为流动相,建立了测定条件。绘制标准曲线得回归方程为Y=0.0017X+0.0131,r=0.9975。氟卡胺及内标的回收率均大于94%,日内及日间变异系数均小于5%。正常人口服氟卡胺后的药代动力学参数,Ka=0.908h-1,T1/2K=8.131h,TPK=3.487h,AUC=441.8ng.h/ml。病人口服氟卡胺的有效血浓度为423.64±227.4ng/ml。Abstract: A high performance liquid chromatographic method (ultraviolet detection) for the determination of flecainide concentration in human plasma was reported. Flurazepam (dalmadorm) was used as the internal standard. A mixture of methanol and deionized water containing ion paired reagent served as the mobile phase. The regression equation of flecainide was. Y=0.0017x+ 0.0131,r= 0.9975. The extraction recoveries exceeded 94%, and the coefficients of variation (both within the same day and in different days) were less than 5 %. The pharmacokinetic parameters in Chinese volunteers after taking flecainide were: Ka = 0.908h-1, T1/2 = 8.131h, peats time = 3.487h,AUC=441.8ng · h/ml.The mean plasma concentration of flecainide in cases responded well to this drug was 423.6±227.4ng/ml.
