苯那普利对糖尿病大鼠肾脏细胞凋亡的影响

EFFECTS OF BENAZEPRIL ON APOPTOSIS IN THE KIDNEY OF DIABETIC RATS

  • 摘要: 目的探讨血管紧张素转换酶抑制剂(ACEI)苯那普利对糖尿病大鼠肾脏细胞凋亡及凋亡相关基因表达的影响。方法单侧肾切除大鼠ip STZ诱发糖尿病模型,每日ig苯那普利(10 mg·kg-1),共12周。采用原位末端标记法检测肾脏细胞凋亡情况,流式细胞术和免疫组化检测肾皮质凋亡相关基因Fas和Fas-L的表达。结果糖尿病组较对照组肾小球、肾小管凋亡细胞数明显增多,Fas和Fas-L的表达亦显著增强;苯那普利治疗组较糖尿病组凋亡细胞数减少,Fas和Fas-L的表达减弱。结论苯那普利可能通过影响凋亡相关基因Fas和Fas-L的表达而抑制肾脏细胞凋亡,从而发挥肾脏保护作用。

     

    Abstract: AIM To investigate the effects of angiotensin converting enzyme inhibitor (ACEI) on apoptosis and the expression of Fas and Fas L in the kidney of diabetic rats. METHODS Uninephrectomized Spraque Dawley rats were used to induce diabetes by intraperitoneal injection of streptozotocin (65 mg·kg-1 ). Benazepril (10 mg·kg-1 ) was given daily by gavage from the next day of the induction to diabetes for 12 weeks. Apoptosis was evaluated by means of terminal-deoxynucleotidyl transferase mediated d-UTP nick end labeling ( TUNEL ). Flow cytometry and immunohistochemistry were used to detect the expression of Fas and Fas-L. RESULTS Compared with those in the kidneys of control group, apoptotic cells were more in number and the expression of Fas and Fas-L was higher in the diabetic kidneys ( P <0.05). The number of apoptotic cells and the level of expression of Fas and Fas-L were reduced by benazepril treatment ( P <0.05). CONCLUSION Angiotensin converting enzyme inhibitor benazepril showed some renal protective effect on diabetic nephropathy, party through inhibiting apoptosis by down-regulating Fas and Fas-L expression.
