Abstract:
Sophoridine in rabbit plasma was determined by gas chromatography. The flow rates of nitrogen, hydrogen and air were 60, 40 and 380 ml/min, respectively. Working temperatures were: column oven 190℃, injection oven 280℃, detector oven 300℃. Sophoramine was used as internal standard. The recovery of sophoridine from plasma was 103.65±2.4% (CV 2.34%) at concentration 3.0μg/ml and 101.55±6.5% (CV 6.35%) at 10.0μg/ml. There was a linear relation between the ratio of peak area and plasma concentration in the range of 1 to 15μg/ml. Detection limit was 0.4μg/ml. The CV values estimated within day and on day-to-day at different concentrations were found to be less than 6%. The pharmacokinetics of sophoridine in rabbits were fitted by an open two compartment model. The composite parameters following iv injection of 20 mg/kg were: α, 0.0864 min
