青藤碱的药理作用Ⅳ.青藤碱降压机制的研究

THE MECHANISM OF HYPOTENSIVE ACTION OF SINOMENINE

  • 摘要: 给麻醉狗、大白鼠和兔静脉注射青藤碱盐酸盐后,皆可产生肯定的降压作用.(?)只狗注射青藤碱(1毫克/公斤)后,平均降压面积为-40±3%.应用同量的药物反复注射三种动物都能出现快速耐受现象;以狗最为明显.在位狗心实验中发现青藤碱在降压同时并不抑制心脏,抗组织胺药不能影响其降压效果,对于M-胆碱反应系统并无影响.本药具抗肾上腺素作用和神经节阻断作用,且能抑制多种中枢性加压反射,故其降压机制可能系抗肾上腺素、阻断神经节和中枢作用的结果.

     

    Abstract: Intravenous injection of Sinomenine hydrochloride to anaesthetized animals (dogs, rats, and rabbits) produced apparent maximal hypotensive effects. In dogs (1 mg/kg), the latent period of hypotension was 22±2.3 seconds; the hypotensive area was -40±3% and the action lasted 25-100 minutes. Repeated administrations revealed tachyphylaxis. The same results were obtained in rats, but the fall of arterial pressure was only transient in rabbits. Our experiments indicate that Sinomenine apparently inhibits the pressor responses induced by the occlusion of the common carotid arteries and the stimulation of the cen- tral ends of sciatic or vagus nerves. It inhibits the effects of stimulation of the distal end of vagus nerve, the effects of excitment of respiration, and the pressor effect by lobeline. Sinomenine also reduces the hypotensive actions of adrenaline and noradrena- line. The hypotensive action of Sinomenine is not due to histamine liberation. Sino- menine exerts no influence on the M-cholinergic reaction system or the cardiac depres- sion. Therefore it is suggested that the hypotensive effects of Sinomenine are mainly due to the central inhibition, the adrenolytic effects, and probably also, the ganglionic block- ing action.
