高分辨率熔解曲线技术及其在中药DNA分子鉴定中的应用

High resolution melting and its application in identity study of traditional Chinese medicine

  • 摘要: 高分辨率熔解曲线(high resolution melting, HRM)是建立在熔解曲线分析的基础上,通过一类新型饱和荧光染料和更高分辨率的检测仪器,主要应用于突变扫描、基因分型和序列匹配等的一项新技术。该技术因简便、精确、快速、闭管、低成本、高通量、对样本无污染等优点而受到广泛关注。本文对高分辨率熔解曲线的原理、影响因素、优势、不足及在中药材HRM分析应用现状等进行系统总结,并对其在中药分子鉴定中的应用前景进行展望。

     

    Abstract: High resolution melting(HRM), based on melting curve analysis, requires not only saturating dyes that fluoresce in the presence of double-stranded DNA, but also higher resolution detection equipment. The melting curve is a novel method for sequence matching, genotyping and mutation scanning. The technology is simple, accurate, rapid, closed-tube, low-cost, and high-throughput, which make it gain more and more applications. This review article presents the basic principles, key factors and both the advantage and limitations of HRM. The potential application is discussed in the study of molecular identity of traditional Chinese medicine.
