The interaction of β-cyclodextrin with etoposide studied by fluorescence spectroscopy

    采用荧光光谱法研究了β-环糊精 (β-CD) 与依托泊苷 (VP-16) 之间的超分子包结作用。考察了时间、温度和浓度对包结反应的影响。并用紫外-见光光谱法对包结物溶液进行了表征。实验结果表明: VP-16本身具有天然荧光, β-CD形成11的超分子包结物后, 荧光强度增大, β-CDVP-16有荧光增敏作用。在22 ℃,  pH 7.0, 包结常数为2.63×105 L·mol−1。根据VP-16316 nm处发射强荧光, 建立了定量检测微量VP-16的荧光分析法, 其线性良好, 相关系数为0.999 9

     

    Abstract:

    The fluorescence spectroscopy and UV spectroscopy have been used to monitor the inclusion  phenomena of VP-16 with β-cyclodextrin (β-CD), together with studies concerning the effects of reaction time, temperature and concentration on this behavior.  The results show that the fluorescence intensity increased when VP-16 and β-CD forming a 11 inclusion complex, which indicate that β-CD has fluorescence sensitizing effect on the VP-16.  At 22 , the inclusion constant was 2.63×105 L·mol−1 at pH 7.0. VP-16 has maximum emission wavelength at 316 nm under the optimum conditions.  According to this, the quantitative micro-detection method of VP-16 by fluorescence spectrometry was established.  The linear regression equation was y = 1.107 89 × 107x + 95.898 1, with a correlation coefficient of 0.999 9.  The detection limit was 2.094×10−7 mol·L−1.
