抗肿瘤抗生素C1027对人结肠癌HCT 8细胞凋亡相关基因表达的影响

摘要: 目的:研究抗肿瘤抗生素C1027 对人结肠癌HCT-8 细胞凋亡相关基因表达的影响。方法:利用cDNA 排列(cDNAarrays)、Northern 杂交、RT-PCR 检测技术。结果:用10 nmol·L-1 C1027 处理HCT-8 细胞8 h 后,利用cDNA 排列检测,发现C1027 可以促进TRIP,TRAF3 ,DR4 ,MCH6 ,MCH4,Apo-3 ,DR5,ABLL,STAT1 等基因的表达,抑制rhoC的表达。用Northern 杂交及RT-PCR 也得到了同样结果。结论:C1027 可能通过调节TNF 受体家族有关的凋亡信号传导通路,诱导细胞凋亡,发挥抗瘤活性。Abstract: AIM: To study the effcet of C1027 on the expression of genes related to apoptosis in HCT-8 human colon cancer cells. METHODS: HCT-8 human colon cancer cells were treated with C1027(10 nmol·L-1) for 8 hours. The effect of C1027 on the expression of genes in HCT-8 cells were detected by cDNA arrays, Northern blot and RT-PCR. RESULTS: Hybridization of the entire cDNA populations to Atlas Arrays showed that C1027 up regulated the expression levels of TRAF3, DR4, DR5, MCH4, MCH6, TRIP, Apo-3, ABLL and STAT1 and down regulated the expression levels of rhoC. These changes were confirmed by Northern blot and RT-PCR. CONCLUSION: Most of the genes are involved in TNF receptor family related signal transduction. The results indicate that C1027 may induce apoptosis by regulating TNF receptor family related signal transduction.
