7-溴乙氧苯四氢巴马汀对离体豚鼠工作心脏的作用

EFFECTS OF 7- BROMOETHOXYBENZENE-TETRAHYDROPALMATINE ON ISOLATED WORKING GUINEA PIG HEART

  • 摘要: 用Tris-丙酮酸钠液灌流离体豚鼠工作心脏,记录左室压的导管从左房灌流管插入,可使心脏有效工作时间达70min。用Tris-丙酮酸钠液加0.5%的氟碳液能使有效工作时间延长至90min。7-溴乙氧苯四氢巴马汀(7-bromoethyoxybenzene tetrahydropalmatine,EBP)及哌唑嗪0.1μmol/L对工作心脏各项指标均无显著影响。甲氧胺1μmol/L,多巴胺1μmol/L对LVP,ABF,T-CO等指标均有明显的药理作用。EBP 10μmol/L能对抗甲氧胺、多巴胺对上述指标的影响。

     

    Abstract: The effective period of isolated working guinea pig heart perfused with Tris-sodium-pyruvate solution(Tris-SP)was shown to be 70 rain. Perfluorotributylamine at a concentration of 0.5% added to Tris-SP prolonged the effective time to 90 min. 7-Bromoethyoxybenzene-tetrahydropalmatine (EBP) 10 μmol/L,and prazosin 0.1 μmol/L showed no obvious influence on the parameters in the isolated working guinea pig heart. Methoxamine 1 μmol/L and dopamine 1 /μmol/L produced marked changes on LVP, ABF, +dp/dtmax and T-CO. It is suggested that the a1-adrenoceptor was partially associated with the positive chronotropic action of the heart. Effects of methoxamine and dopamine on the parameters were antagonized by EBP.It shows that the antagonistic effects of EBP on methoxamine and dopamine are concerned with its antagonistic effection on a1-adrenoceptor and dopamine reccptor This is consistent with previous results in rabbit aortic strips and tat anococcygeus muscles.
