力达霉素对人结肠癌HCT-8细胞侵袭调节基因表达的影响

EFFECT OF LIDAMYCIN ON THE EXPRESSION OF GENES INVOLVED IN INVASION REGULATION IN HCT-8 HUMAN COLON CANCER CELLS

  • 摘要: 目的研究抗肿瘤抗生素力达霉素对人结肠癌HCT-8细胞侵袭调节基因表达的影响。方法利用cDNA排列(cDNAarrays)、Northern杂交、RT-PCR技术。结果力达霉素可以促进TIMP-1的表达,抑制MMP-9的表达,cDNA排列、Northern杂交、RT-PCR检测得到了同样的结果。结论力达霉素可能通过抑制IV型胶原酶的产生,同时诱导金属蛋白酶抑制因子的产生,从而表现出抗侵袭活性。

     

    Abstract: AIM To study the effect of lidamycin on the expression of genes involved in invasion regulation in HCT-8 human colon cancer cells. METHODS HCT-8 human colon cancer cells were treated with lidamycin (10 nmol·L-1) for 8 h. The effect of lidamycin on the expression of genes were detected by cDNA arrays, Northern blot and RT-PCR. RESULTS Hybridization of the entire cDNA populations to Atlas Arrays showed that lidamycin down-regulated the expression level of MMP-9 and up-regulated the expression level of TIMP-1. These changes were confirmed by Northern blot and RT-PCR. CONCLUSION The results indicate that lidamycin may exhibit its anti-invasive activity by inhibitting the production of type IV collagenase whilst enhancing the production of tissue inhibitor metalloproteinase.
