三尖杉生物碱的毛细管气相色谱分析

GLASS CAPILLARY GAS CHROMATOGRAPHY OF CEPHALOTAXUS ALKALOIDS

  • 摘要: OV-17SCOT玻璃毛细管柱在恒温条件下,除能分离有抗肿瘤活性的酯碱类化合物外,还能分离填充柱不能分离的11-羟基三尖杉碱和脱甲基三尖杉酮碱。分析了不同产地的三尖杉植物提取物中的总生物碱,用毛细管气相色谱和质谱联用鉴定了从A产地得到的总生物碱中的4个峰,其中一个可能属于酯碱类型的新生物碱。并测定了混合酯碱的含量,结果表明目前采用的紫外光谱法误差太大,应提高对杂质检查的要求或经薄板层析分离后,再测定含量。

     

    Abstract: In addition to the antitumor-active esters, 11-hydroxycephalotaxine and desmethylcephalotaxinone which were not resolved on packed column were successfully separated with SCOT glass capillary coated with OV-17 at 268℃. The crude alkaloid mixtures extracted from Cephalotaxus plants from four regions were analysed, and(GC)2-MS was used to identify four components in the crude alkaloid mixtures extracted from Cephalotaxus plant of region A, one of them seemed to be a newcompound of Cephalotaxine ester type. The quantitative glass Capillary gas chromatography of the mixtures of harringtonine and homoharringtonine showed that there was a great error in direct UV spectrometry.
