多波长高效液相色谱法同时测定栀子中的三类成分

Simultaneous determination of 3 kinds of components in Gardenia by high-performance liquid chromatography under different UV-vis wave length

  • 摘要: 目的同时测定栀子中3类有效组分,9个成分(栀子酸、栀子苷、异栀子苷、京尼平龙胆二糖苷、绿原酸、藏红花酸、3种藏红花素)的含量。方法紫外-可见三波长同时检测的高效液相色谱法:色谱柱为Phenomenex Luna C18柱(250 mm×4.6 mm ID,5 μm);流动相为(A)0.3%甲酸水溶液, (B)甲醇-乙腈(9∶1);流速为0.8 mL·min-1;柱温为35 ℃;检测波长为 240 nm(栀子酸、栀子苷、异栀子苷、京尼平龙胆二糖苷), 330 nm(绿原酸)和440 nm(藏红花酸和3种藏红花素);线性梯度洗脱。结果建立了同时对栀子中的9个成分进行定量的测定方法,并将此方法用于5个产地栀子各成分的测定。结论为中药材栀子提供了更合理、可靠的质控方法。

     

    Abstract: AimTo determine 3 kinds of biologically active components (9 compounds) in Gardenia, namely geniposidic acid, gentiobioside, gardenoside, genipin geniposide, chlorogenic acid, crocin 1, crocin 2, crocin 3 and crocetin simultaneously and quantitively. MethodsAn HPLC method detected by 3 different UV-vis waves — 240 nm for geniposidic acid, geniposide, gardenoside, genipin gentiobioside,330 nm for chlorogenic acid, 440 nm for crocin 1, crocin 2, crocin 3 and crocetin respectively, has been developed: Phenomenex Luna C18 column(250 mm×4.6 mm ID,5 μm)with linear gradient elution using methanol-acetotrile(9∶1) and 0.3% aqueous formic acid. ResultsThis method successfully determined 9 compounds in Gardenia (from 5 different areas) simultaneously and quantitively. ConclusionThis is a more reasonable and credible quality control method for the Chinese traditional medicine.
