醋酸泼尼松对大鼠松质骨和密质骨影响的比较

SKELETAL EFFECTS OF PREDNISONE ACETATE ON TRABECULAR AND CORTICAL BONE IN SPRAGUEDAWLEY RATS

  • 摘要: 目的:比较醋酸泼尼松对大鼠胫骨上段和腰椎松质骨及胫骨中段密质骨的影响。方法:醋酸泼尼松45mg·kg-1,ig,1周2次共90d。骨标本行不脱钙骨制片,骨组织形态计量学测量。结果:松质骨:激素组与对照组比,胫骨上段(PTM)和腰椎(LV)松质骨的骨量减少,骨结构变差;代表骨形成的参数值降低,而骨吸收参数值增加。与自身对照组比,PTM骨量减少40%,LV骨量减少28%。密质骨:激素组与对照组比,实验90d骨量未出现减少。骨外膜面代表骨形成的参数减少,骨内膜骨吸收参数增加。结论:醋酸泼尼松以抑制骨形成、促进骨吸收而致骨丢失。

     

    Abstract: AIM: To study the effects of prednisone acetate on the histomorphology of rat skeleton. METHODS: 3Monthold male rats were given prednisone acetate (4.5mg·kg-1) ig, twice a week for 90 days. The proximal tibial metaphysis (PTM), lumber vertebra (LV) and the middle part of tibial shafts (Tx) were processed undecalcified for quantitive bone histomorphometry. RESULTS: PTM: prednisone group vs aging group, trabecular bone loss and poor architecture were found in prednisone acetate treated group. Bone formation indices were decreased and bone resorption perimeter was increased in prednisone group. The change of LV was almost the same as PTM, but the change was less than PTM. TX: The cortical bone and marrow areas of tibial shaft did not change in either the aging control group or the prednisone acetate group during the 90 day experimental period. A decrease in bone formation indices in periosteal surface of Tx in prednisone acetate group was observed. Prednisone acetate markedly increased the percentage of eroded perimeter in the endocortical surface compared with the control group. CONCLUSION: The results show that prednisone acetate has significant effect on the proximal tibial metaphysis, lumber vertebra and the middle part of tibial shafts. These indicate that different part of the bone have different reaction to prednisone acetate.
