SYNTHESIS OF β-DIMETHYLAMINOETHYL DIPHENYL-PROPYLACETATE HYDROCHLORIDE (SKF 525 A)

  • 摘要: β-二乙胺基乙醇二苯基丙基乙酸酯盐酸盐(SKF 525 A)为药物转化酶研究用的试剂。所需的二苯基丙基乙酸,系由下法制得:安息香与丙基格氏试剂反应可得二醇,用酸重排可得醛,再用高锰酸钾氧化可得酸。

     

    Abstract: The title compound is of value in the studies of drug-metabolizing enzymes.The following steps were devised to obtain the acid moiety, diphenylpropylacetic acid.Propylmagnesium chloride was allowed to react with benzoin to give a glycol, the latter rearranging under acidic conditions to give an aldehyde. Permanganate oxidation of the aldehyde gave the desired acid.
