PROTECTION AND ANTI-OXIDATIVE MECHANISM OF 2(3)-TERT-BUTYL-4-HYDROXYANISOLE AGAINST ETHANOL INDUCED GASTRIC LESION IN RATS

  • 摘要: 目的:研究叔丁基-4-羟基茴香醚(BHA)对乙醇诱发胃粘膜损伤的保护作用。方法:通过给大鼠ig无水乙醇制备胃粘膜损伤模型,测定胃粘膜内一些酶的活性和MDA含量。结果:ig BHA 20和100 mg.kg-1.d-1连续3 d,能显著防止乙醇诱发的胃粘膜损伤,并降低由乙醇所致的MDA含量增加。BHA有诱导醌还原酶、谷胱甘肽还原酶活性增加的作用,且能阻止由乙醇引起的醌还原酶、谷胱甘肽还原酶和SOD活性的降低。结论:BHA对乙醇诱发大鼠胃粘膜损伤有保护作用,其作用机制可能与BHA诱导抗氧化酶活性增高和抗脂质过氧化作用有关。

     

    Abstract: AIM: The protection and anti-oxidative mechanism of 2(3)-tert-butyl-4-hydroxyanisole(BHA) against ethanol-induced gastric mucosa lesion in rats were evaluated. METHODS: Gastric mucosal lesions were induced by oral administration of absolute ethanol in rats. The activities of some enzymes and MDA content in gastric mucosa were determined. RESULTS: Oral administration of BHA at 100 mg.kg-1 and 20 mg.kg-1 reduced significantly the gastric lesion induced by ethanol, and inhibited the increase of MDA in gastric mucosa. Compared with normal control, BHA was found to increase the activities of quinone reductase (QR) and glutathione reductase (GR). In ethanol-treated rats decreases of QR, GR and SOD were observed. However, BHA was shown to prevent the decrease of QR, GR and SOD in gastric mucosa induced by ethanol. CONCLUSION: These results suggest that BHA has protective effect against ethanol-induced gastric mucosal lesion via increasing the activities of anti-oxidative enzymes and anti-lipid peroxidation.
