Abstract:
The present paper reports the central and peripheral actions of d1-3-n-butyl-phthalide in animals. Beside the anticonvulsant action, Ag-1 was also found to be able to protect mice and rats from seizures induced by sodium glutamate, spasm induecd by i.c.v, acetycholine and tremorine spasm as well as electroshock. No change of EEG patterns was shown in 13 unanesthetized normal rabbits after ip injection of Ag-1 50, 120, 200 and 250 mg/kg. After a single iv of metrazol 20 mg/kg to rabbits, bursts of spike waves were recorded, associated with violent general convulsions. When Ag-1 was injected 30 min prior to metrazol, a similar effect on EEG patterns was recorded, but no behavioral seizure was observed.This compound was also found to have a direct action on acetylcholine or barium chloride induced spasm of isolated rat ileum. Ag-1 in the concentration of 9.5×10
-7M was shown to be able to antagonize the spasmodic action of acetylcholine (2×10
-9 M) and barium chloride (2×10
-6 M) on the rat ileum.
