瑞香素与中枢单胺类介质的关系

THE RELATIONSHIP BETWEEN DAPHNETIN ANALGESIA AND CENTRAL NEUROTRANSMITTERS OF THE MONOAMINES

  • 摘要: 本文报道从中草药祖师麻中提得的单一成分瑞香素(7,8-二羟基香豆素),也曾称为祖师麻甲素。其镇痛作用能被5-羟色胺(5-HT)和去甲肾上腺素(NA)所加强,同时本实验室测定了在药物作用的峰值时间几个脑区的单胺类介质(5-HT,NA,DA)及代谢物(5-HIAA)的含量,结果显示药物使“低位脑干”(包括中脑、桥脑和延脑)部位的5-HIAA含量显著增加(+133.3%),而对其它脑区及介质均无明显作用。

     

    Abstract: Daphnetin found in the Chinese herb Daphne giraldii Nitsche has been used as a non-narcotic analgesic in the clinic.When injected intraperitoneally in mice (100 mg/kg), its analgesic effects were potentiated by intraventricular injection of serotonin (5-HT, 20μg) or naradrenaline (NA, 1.25μg).The effect of daphnetin on the concentrations of noradrenaline (NA), dopamine (DA), 5-hydroxytryptamine (5-HT) and 5-hydroxyindoleacetic acid (5-HIAA), which are found in end-brain, interbrain and lower brain stem (including mid-brain, pons, medullary brain) in rats were determined.Experimental results indicate that daphnetin markedly increased the 5-HIAA content (133.3%) but did not affect the 5-HT content in lower brain stem. No other statistically significant changes of neurotransmitter had been observed.
