禾叶山麦冬中一个新的C27甾体苷硫酸酯类化合物

A novel C27-steroidal glycoside sulfate from Liriope graminifolia

    为了对禾叶山麦冬 (Liriope graminifolia (Linn.) Baker) 进行植物化学和药理活性的研究, 应用现代色谱技术对禾叶山麦冬植物的化学成分进行了提取分离, 应用HR-MS1H NMR13C NMR1H-1H COSYHSQCHMBCNOESY技术对分离得到的4个化合物进行了结构确证。4个化合物分别鉴定为: (25S)-ruscogenin 1-sulfate-3-O-α-L-rhamnopyranoside (1)(25S)-ruscogenin 1-O-β-D-xylopyranosyl-3-O-α-L-rhamnopyranoside (2)橙皮苷 (hesperidin, 3) 4', 7-二羟基-5-甲氧基二氢黄酮 (4)。其中化合物1为一个罕见的具有27个碳原子骨架结构的新的甾体苷硫酸酯类化合物, 并对K562细胞及HL60细胞具有显著抑制作用。

     

    Abstract:

    An unusual novel C27-steroidal glycoside sulfate was isolated from the underground organs of Liriope graminifolia (Linn.) Baker with three known compounds.  Their chemical structures were determined by spectral analysis, including HR-MS, 1D and 2D NMR as (25S)-ruscogenin 1-sulfate-3-O-α-L-rhamnopyranoside (1), (25S)-ruscogenin 1-O-β-D-xylopyranosyl-3-O-α-L-rhamnopyranoside (2), hesperidin (3), and 4', 7-dihydroxy- 5-methoxyflavanone (4).  Compound 1 has cytotoxic activities against K562 and HL60 cells with IC50 values of 18.6 μg·mL−1 and 16.5 μg·mL−1, respectively.
