A NEW CHROMATOGRAPHY SYSTEM FOR SEPARATION OF BIOMOLECULES

  • 摘要: 本文介绍了一种较新的分离蛋白质类的快速方法,即快速蛋白液相层析(FPLC)法。采用对蛋白质有高效能的亲水性聚醚所组成的凝胶,在中等压力下达到快速高效分离生物分子如蛋白质、酶、多肽及多核苷酸等。作者介绍了FPLC仪器及实验条件。用此法快速、简便、适于分离肾衰竭病人尿中血清蛋白,可应用于临床研究。

     

    Abstract: In this paper we have described a new methodology for fast separation of charged physiological biopolymers with retained or improved resolution. Besides being of great interest in fields where ionexchange chromatography traditionally are used, the speed and simplicity of the FPLC technique offers interesting, possibilities in clinical research. On basis of the results obtained FPLC seems to be a suitable technique for separating serum proteins in urine. The protein compositions of urines from different pathophysiological states differ considerably from each other and from normal urines indicating that this technique could be used for rapid monitoring of the status of patients with various diseases.
