刺参酸性粘多糖的分离及其理化性质

AN ACIDIC MUCOPOLYSACCHARIDE ISOLATED FROM STICHOPUS JAPONICUS SELENKA AND SOME OF ITS PHYSICAL AND CHEMICAL PROPERTIES

  • 摘要: 刺参为一海洋动物性药材,具有多种医疗用途。作者以刺参体壁(中药药用部位)经胃蛋白酶及胰匀浆水解去蛋白,以乙醇沉淀粗多糖,后者采用氧化法脱色及二乙氨乙基纤维素分离、提纯,获得具有抗肿瘤活性的刺参多糖。本品显示强异染活性,电泳及超离心分析证明为单一成份。对多糖的组成分析指出,本品含氨基半乳糖,已糖醛酸、岩藻糖和硫酸酯,其分子比值为1∶1∶1∶4,可以认为本品为一种硫酸粘多糖。该多糖在组成上与文献报道的各种海参多糖不同。本研究测定了刺参酸性粘多糖的比旋度、特性粘度及沉降系数。

     

    Abstract: Stichopus japonicus Selenka, a kind of sea cucumber, has long been used in traditional Chinese medicine. An antitumor polysaccharide was extracted by hydrolysing its cell wall with both pepsin and pancreas homogenate followed by fractional precipitation with alcohol. After decolorization, the crude product was further purified by chromatography on a DEAE-cellulose column. The polysaccharide was obtained in acetate buffers (pH 6.0)containing 0.8 M NaGl0 as eluate. By electrophoretic and ultracentrifugal analysis, the preparation was shown to be quite pure. The specific rotation, intrinsic viscosity and sedimentation constant were measured, and a high metachromatic activity was demonstrated. The component sugars of the polysaccharide were identified as. galactosamine, hexuronic acid, fucose and sulfate, and their molar ratio was about 1:1: 1: 4, respectively. Thus the sulfated mucopolysaccharide obtained is widely different from all the known acidic polysaccharides isolated previously from sea cucumbers including stichopus iaponicus.
