鞘脂在肝脏疾病中的研究进展

Recent advances in study of sphingolipids on liver diseases

  • 摘要: 鞘脂是生物膜的结构组分。近年来研究发现,鞘脂分子(特别是神经酰胺、1-磷酸鞘氨醇)亦具有重要的生物活性,参与细胞增殖、分化、基因表达及凋亡等生理过程。越来越多的研究表明,鞘脂参与了肝脏疾病的发生发展过程,如肝脏缺血再灌注损伤、脂肪肝、肝细胞癌等。也有研究表明,细胞内特定鞘脂的堆积是诱发肝功能障碍的重要因素。因此,调控鞘脂代谢通路可能成为治疗肝脏疾病的潜在靶点。本文综述了鞘脂代谢通路及其在肝脏疾病中的作用及鞘磷脂代谢酶相关药物的研究进展。

     

    Abstract: Sphingolipids, especially ceramide and S1P, are structural components of biological membranes and bioactive molecules which participate in diverse cellular activities such as cell division, differentiation, gene expression and apoptosis. Emerging evidence demonstrates the role of sphingolipids in hepatocellular death, which contributes to the progression of several liver diseases including ischaemia-reperfusion liver injury, steatohepatitis or hepatocarcinogenesis. Furthermore, some data indicate that the accumulation of some sphingolipids contributes to the hepatic dysfunctions. Hence, understanding of sphingolipid may open up a novel therapeutic avenue to liver diseases. This review focuses on the progress in the sphingolipid metabolic pathway with a focus on hepatic diseases and drugs targeting the sphingolipid pathway.
