黄芪多糖的研究

STUDIES ON THE POLYSACCHARIDES OF “HUANG QI” (ASTRAGALUS MONGHOLICUS BUNGE.)

  • 摘要: 从中药黄芪Aslragalus mongholicus Bunge.的水提液中分离得到两种葡聚糖AG-1、AG-2和两种杂多糖AH-1、AH-2。该四种多糖经电泳、凝胶柱层析等证实均为单一组分。AG-1为水溶性,经水解、过碘酸氧化、甲酸生成、Smith降解、乙酰解、红外、C-13核磁共振等确定为α(1→4)(1→6)葡聚糖、α(1→4)与口(1→6)甙键糖基的组成比例约为5:2。AG-2不溶于水,为α(1→4)葡聚糖。AH-1为水溶性酸性多糖,水解后纸层析检出含己糖醛酸(半乳糖醛酸和葡萄糖醛酸)、葡萄糖、鼠李糖、阿拉伯糖,其分子比值为1:0.04:0.02:0.01。AH-2为水溶性、水解后用纸层析和气相层析检出葡萄糖和阿拉伯糖,其分子比值为1:0.15。AG-1、AH-1具有某些免疫促进作用。

     

    Abstract: Two glacans AG-1, AG-2 and two heterosaccharides AH-1, AH-2 were isolated and purified from water extract of Chinese drug "Huang Qi".By electrophoretic and gel chromatography, the four polysaccfiarides were shown to be single substances.According to IR data and acid hydrolysis, AG-1 was identified as a α-glucan. Result of periodate oxidation, Smith type degradation, acetolysis and C-13 NMR showed that the glucan has a structure as α-(1→4) (1→6)glucan. The ratio of α-(1→4) and α-(1→6) linkage was about 5∶2.AG-2 was insoluble in water, on the basis of IR, acid hydrolysis, periodate oxidation, acetolysis and C-13 NMR estimation, it was elucidated as α-(1→4) glucan.AH-1 was an acidic polysaccharide, the component sugars after hydrolysis of the polysaccharide were identified as hexouronic acid (galacturonic acid and glucuronic acid), glucose, rhamnose, and arabinose, their molecular ratio was about 1∶0.04∶0.02∶0.01.AH-2 was soluble in water, the result obtained from acid hydrolysis and identified by PC, GC, the polysaccharide composed of glucose and arabinose, the molecular ratio of these sugars was 1∶0.15.AG-1 and AH-1 showed some activity in immunological study.
